From 128 North or South:

Take exit 31B. Go approximately 1/4 mile and bear right into the jug-handle turn (which will allow you to make a U-turn on Bedford Street). Stay in the left lane of the jug handle. Take a left at the light. The entrance to the Boston Children's Hospital at Lexington is the first driveway on the right after the turn.

Children's Hospital at Lexington is easily reachable by public transportation. Take the MBTA Red Line train to Alewife then take the #62 (Bedford V.A. Hospital - Alewife Station) MBTA Bus route. The bus will drop you off at the entrance of the building.
